Th2 Cytokine Expression in Mouse Splenocytes Infected with Echinostoma hortense Crude Extracts

Abstract

|
Echinostoma hortense is a common intestinal parasite in South-East Asia where it has high prevalence. But underlying mechanism implicated with clinical manifestation or immune reaction is unknown. To clarify immune status induced by E. hortense crude extracts in BALB/c mouse, Th2 cytokine(IL-4 and IL-5) profiles are examined by RT-PCR and ELISA. 6 week-old female BALB/c mouse is used to obtain splenocytes. This splenocytes are examined for the Th2 cytokine expression after treating with E. hortense crude extracts. The result of Th2 cytokine mRNA expression using RT-PCR, IL-4 expression is peak after 1 hours and continues to 6 hours, and IL-5 expression is peak after 1 hour and continues to 12 hours. Also Th2 cytokine production using ELISA, IL-4 cytokine is more produced in 9 hours. In conclusion, E. hortense crude extracts might have play the role of stimulator to mouse splenocytes and IL-4 is more produced than I1-5 during stimulation with crude extracts.
